Abstract: The transcription factors DREBs/CBFs specifically interact with the dehydration-responsive element/C-repeat ( DRE/CRT ) cis-acting element ( core motif : G/ACCGAC ) and control the expression of many stress-inducible genes in Arabidopsis . In rice , we isolated five cDNAs for DREB homologs : OsDREB1A , OsDREB1B , OsDREB1C , OsDREB1D , and OsDREB2A . Expression of OsDREB1A and OsDREB1B was induced by cold , whereas expression of OsDREB2A was induced by dehydration and high-salt stresses . The OsDREB1A and OsDREB2A proteins specifically bound to DRE and activated the transcription of the GUS reporter gene driven by DRE in rice protoplasts . Over-expression of OsDREB1A in transgenic Arabidopsis induced over-expression of target stress-inducible genes of Arabidopsis DREB1A resulting in plants with higher tolerance to drought , high-salt , and freezing stresses . This indicated that OsDREB1A has functional similarity to DREB1A . However , in microarray and RNA blot analyses , some stress-inducible target genes of the DREB1A proteins that have only ACCGAC as DRE were not over-expressed in the OsDREB1A transgenic Arabidopsis . The OsDREB1A protein bound to GCCGAC more preferentially than to ACCGAC whereas the DREB1A proteins bound to both GCCGAC and ACCGAC efficiently . The structures of DREB1-type ERF/AP2 domains in monocots are closely related to each other as compared with that in the dicots . OsDREB1A is potentially useful for producing transgenic monocots that are tolerant to drought , high-salt , and/or cold stresses .

Abstract: One of the major environmental factors limiting plant productivity is lack of water . Abstract: Rice yield is greatly affected by environmental stresses such as drought and salinity . In response to the challenge of producing rice plants tolerant to these stresses , we introduced cDNA encoding the transcription factors DREB1A and DREB1B under the control of the stress inducible rd29 promoter . Two different indica rice cultivars were used , BR29 , an improved commercially cultivated variety from Bangladesh and IR68899B , an IRRI bred maintainer line for hybrid rice . Agrobacterium mediated transformation of BR29 was done independently with DREB1A isolated from rice and Arabidopsis and DREB1B isolated from rice , whereas biolistic transformation was done with rice DREB1B in the case of IR68899B . Initial genetic integration was confirmed by PCR and Southern blot analysis . Salinity tolerance was assayed in very young seedlings . Drought stress tests were found to be more reliable when they were carried out at the pre-flowering booting stage . RNA gel blot analysis as well as quantitative PCR analysis was performed to estimate the transcription level under stressed and unstressed conditions . Agronomic performance studies were done with stressed and unstressed plants to compare the yield losses due to dehydration and salt loading stresses . Noticeably enhanced tolerance to dehydration was observed in the plants transformed with DREB1A isolated from Arabidopsis while DREB1B was found to be more effective for salt tolerance .

Abstract: We show here that transgenic Arabidopsis plants that expressed chimeric repressors derived from the AtMYB102 , ANAC047 , HRS1 , ZAT6 and AtERF5 transcription factors were tolerant to treatment with 400 mm NaCl , which was lethal to wild-type plants . The transgenic plants grew well , without any apparent differences from the wild-type plants under normal growth condition . The transgenic lines expressing the AtMYB102 , ANAC047 and HRS1 chimeric repressors germinated in the presence of 225 mm NaCl , while those expressing the ZAT6 and AtERF5 did not . However , the latter lines were tolerant to osmotic stress and germinated in the presence of 600 mm mannitol , suggesting a link between responses to salt and osmotic stress . Expression of the AtMYB102 , ANAC047 , ZAT6 and AtERF5 genes was induced by salt treatment , while that of HRS1 was repressed . HRS1 has transcriptional repressive activity and appears to suppress the expression of factors that negatively regulate salt tolerance . Microarray analysis revealed that the levels of expression of DREB1A , DREB2B and several genes for ZAT transcription factors rose 10 to 100-fold in the AtMYB102 chimeric repressor line under both normal and stress conditions . Elevated expression of DREB and ZAT related genes might be involved in the salt tolerance of the AtMYB102 chimeric repressor line . Transgenic rice plants expressing chimeric repressors derived from Os02g0325600 and Os03g0327800 , rice homologues of HRS1 and ANAC047 , were tolerant to salinity stress demonstrated by suppression of growth inhibition and ion leakages . Expression of a chimeric repressor provides an effective strategy for enhancing tolerance of plants to abiotic stress .
