Abstract: OBJECTIVE : To determine the effect of Tanshinone IIA ( TanIIA ) on the phosphory-lated NMDA receptor 1 at Serine 897 site ( phospho-NR1 S897 ) and intracellular free calcium concentration ( [ Ca ( 2+ ) ] ( i ) ) in neonatal SD rats with hypoxic ischemic brain damage ( HIBD ) , and to explore the neuroprotective mechanism of TanIIA in HIBD . METHODS : Neonatal SD rats were randomly divided into a normal control , and an HIBD and TanIIA+HIBD group . Rice-Vannucci method was used for HIBD animal model . Time points were : 3 , 6 , 12 , and 24 h after HIBD ( n=10 in each group at each time point ) . TanIIA was intraperitoneally given at 1 mug/g every 12 h . Fura-2AM was used to mark the fluorescent calcium probe and [ Ca ( 2+ ) ] ( i ) was measured by a Hitachi F-4500 Fluorescence Spectrophometer . Fluorescent immunohisotichemical study was used for the expression of phospho-NR1 S897 . RESULTS : ( 1 ) Compared with the normal control group , both the [ Ca ( 2+ ) ] ( i ) absolute number and ipsi-/contra-lateral ratio were increased at each time point with statistical significance ( P<0 . 05 ) . Compared with the HIBD group , the [ Ca ( 2+ ) ] ( i ) in the HIBD+ TanIIA group was decreased at each time point . At 24 h after HIBD , the ipsi-/contra-lateral ratio of HIBD+ TanIIA group was 24 . 9% less than that of HIBD group with statistical significance ( P<0 . 05 ) . ( 2 ) In the normal control group , abundant phospho-NR1 S897 positive cells were nicely distributed in the cortex Compared with the normal control group , at each time point , both the absolute number of phospho-NR1 S897 positive cells and the fluorescent intensity of phospho-NR1 S897 in the ipsilateral cortex of the HIBD group were decreased with statistical significance ( P<0 . 05 ) . Compared with the HIBD group , both the absolute number of phospho-NR1 S897 positive cells and the fluorescent intensity of phospho-NR1 S897 in the ipsilateral cortex of HIBD+ TanIIA were increased . There was significant difference at 3 and 12 h after the HIBD ( P<0 . 05 ) . Abstract: In a previous study , we reported the ecological significance of uncultured bacterial group U2 in the rumen . In this study , the involvement of a recently-cultured group U2 bacterium , strain R-25 , in fiber digestion was tested in coculture with the fibrolytic bacterium Fibrobacter succinogenes S85 . Dry matter ( DM ) digestion , growth and metabolites were examined in culture using rice straw as the carbon source . Although strain R-25 did not digest rice straw in monoculture , coculture of strain R-25 and F succinogenes S85 showed enhanced DM digestion compared to that for F succinogenes S85 monoculture ( 36 . 9+/-0 . 6% vs 32 . 8+/-1 . 3% , P < 0 . 05 ) . Growth of strain R-25 and production of the main metabolites , D-lactate ( strain R-25 ) and succinate ( F succinogenes S85 ) , were enhanced in the coculture . Enzyme assay showed increased activities of carboxymethylcellulase and xylanase in coculture of strain R-25 and F succinogenes S85 . Triculture including strain R-25 , F succinogenes S85 and Selenomonas ruminantium S137 showed a further increase in DM digestion ( 41 . 8+/-0 . 8% , P < 0 . 05 ) with a concomitant increase of propionate , produced from the conversion of D-lactate and succinate . Abstract: The nucleotide sequence of DNA complementary to rice ragged stunt oryzavirus ( RRSV ) genome segment 8 ( S8 ) of an isolate from Thailand was determined . RRSVS8 is 1914 bp in size and contains a single large open reading frame ( ORF ) spanning nucleotides 23 to 1810 which is capable of encoding a protein of M ( r ) 67 , 348 . The N-terminal amino acid sequence of a approximately 43K virion polypeptide matched to that inferred for an internal region of the S8 coding sequence . These data suggest that the 43K protein is encoded by S8 and is derived by a proteolytic cleavage . Predicted polypeptide sizes from this possible cleavage of S8 protein are 26K and 42K . Polyclonal antibodies raised against a maltose binding protein ( MBP ) -S8 fusion polypeptide ( expressed in Escherichia coli ) recognised four RRSV particle associated polypeptides of M ( r ) 67K , 46K , 43K and 26K and all except the 26K polypeptide were also highly immunoreactive to polyclonal antibodies raised against purified RRSV particles . Cleavage of the MBP-S8 fusion polypeptide with protease Factor X produced the expected 40K MBP and two polypeptides of apparent M ( r ) 46K and 26K . Antibodies to purified RRSV particles reacted strongly with the intact fusion protein and the 46K cleavage product but weakly to the 26K product . Furthermore , in vitro transcription and translation of the S8 coding region revealed a post-translational self cleavage of the 67K polypeptide to 46K and 26K products . These data indicate that S8 encodes a structural polypeptide , the majority of which is auto-catalytically cleaved to 26K and 46K proteins . The data also suggest that the 26K protein is the self cleaving protease and that the 46K product is further processed or undergoes stable conformational changes to a approximately 43K major capsid protein .

Abstract: The nucleotide sequences of DNAs complementary to the eighth ( S8 ) and the tenth ( S10 ) largest of the 12 genome segments of rice gall dwarf virus ( RGDV ) were determined . The S8 and S10 segments consist of 1578 and 1198 nucleotides , each with a single open reading frame extending for 1278 nucleotides from nucleotide 21 , and 960 nucleotides from nucleotide 22 , respectively . S8 encodes a polypeptide of 426 amino acids with an Mr of 47419 . The amino acid sequences of several peptide fragments of the major outer capsid protein reported as 45K were contained in the predicted polypeptide . This protein , renamed the 47K protein , showed high homology with the outer capsid proteins of rice dwarf virus ( RDV ) and wound tumour virus ( WTV ) ; there was 56 , 52 and 48% amino acid sequence identity between RGDV and WTV , RGDV and RDV , and RDV and WTV , respectively . S10 had the coding potential for a polypeptide of 320 amino acids with an Mr of 36 , 095 ( 36K protein ) , which exhibits 32% and 35% amino acid sequence identity with the predicted translation product of RDV S9 and the P9 capsid protein encoded by WTV S11 , respectively . The conserved terminal sequences 5 GG . . . GAU 3 which are present in all genome segments of WTV and RDV so far analysed , and in S9 of RGDV , were also found in RGDV S8 and S10 . This conserved sequence together with the segment-specific inverted repeats found in the terminal sequence of RGDV S8 and S10 are thus characteristic structures common to all three phytoreoviruses . Abstract: This is the first sequence-based characterization of Mal de R Cuarto virus ( MRCV ) , currently classified as a variant of Maize rough dwarf virus ( MRDV ) and exclusively found in South America . We sequenced and analyzed genome segments S4 and S8 . MRCV S4 coded for a putative 131 . 67 kDa protein while MRCV S8 coded for a putative 68 . 26 kDa protein containing an ATP/GTP-binding motif . The 5 and 3 ends of MRCV segments , were 5AAGUUUUU3 and 5CAGCUnnnGUC3 , respectively . Prediction of secondary structure of both segments coding strands showed that terminal regions were able to form structures that are proposed to be replication and packaging signals . MRCV S4 showed identity to members of Fijivirus as well as to two other genera of the Reoviridae family . MRCV S8 revealed identity with Rice black streaked dwarf virus ( RBSDV ) S8 , MRDV S7 , Oat sterile dwarf virus ( OSDV ) S9 and Nilaparvata lugens reovirus ( NLRV ) S7 . While MRDV and RBSDV segments are highly homologous between each other , MRCV identity levels with them was considerably lower . We discussed the evolutionary relationships of MRCV to other Reoviridae , and based on phylogenetic analysis we proposed that although MRCV is related to MRDV , it could be regarded as a new species of the Fijivirus genus .
