Abstract: Loss of function of duplicated genes plays an important role in the evolution of postzygotic reproductive isolation . The widespread occurrence of gene duplication followed by rapid loss of function of some of the duplicate gene copies suggests the independent evolution of loss-of-function alleles of duplicate genes in divergent lineages of speciation . Here , we found a novel loss-of-function allele of S27 in the Asian annual wild species Oryza nivara , designated S27-niv ( s ) , that leads to F ( 1 ) pollen sterility in a cross between O sativa and O nivara . Genetic linkage analysis and complementation analysis demonstrated that S27-niv ( s ) lies at the same locus as the previously identified S27 locus and S27-niv ( s ) is a loss-of-function allele of S27 . S27-niv ( s ) is composed of two tandem mitochondrial ribosomal protein L27 genes ( mtRPL27a and mtRPL27b ) , both of which are inactive . The coding and promoter regions of S27-niv ( s ) showed a number of nucleotide differences from the functional S27-T65 ( + ) allele . The structure of S27-niv ( s ) is different from that of a previously identified null S27 allele , S27-glum ( s ) , in the South American wild rice species O glumaepatula , in which mtRPL27a and mtRPL27b are absent . These results show that the mechanisms for loss-of-function of S27-niv ( s ) and S27-glum ( s ) are different . Our results provide experimental evidence that different types of loss-of-function alleles are distributed in geographically and phylogenetically isolated species and represent a potential mechanism for postzygotic isolation in divergent species .

Abstract: In intraspecific crosses between cultivated rice ( Oryza sativa ) subspecies indica and japonica , the hybrid male sterility gene S24 causes the selective abortion of male gametes carrying the japonica allele ( S24-j ) via an allelic interaction in the heterozygous hybrids . In this study , we first examined whether male sterility is due solely to the single locus S24 . An analysis of near-isogenic lines ( NIL-F ( 1 ) ) showed different phenotypes for S24 in different genetic backgrounds . The S24 heterozygote with the japonica genetic background showed male semisterility , but no sterility was found in heterozygotes with the indica background . This result indicates that S24 is regulated epistatically . A QTL analysis of a BC ( 2 ) F ( 1 ) population revealed a novel sterility locus that interacts with S24 and is found on rice chromosome 2 . The locus was named Epistatic Factor for S24 ( EFS ) . Further genetic analyses revealed that S24 causes male sterility when in combination with the homozygous japonica EFS allele ( efs-j ) . The results suggest that efs-j is a recessive sporophytic allele , while the indica allele ( EFS-i ) can dominantly counteract the pollen sterility caused by S24 heterozygosity . In summary , our results demonstrate that an additional epistatic locus is an essential element in the hybrid sterility caused by allelic interaction at a single locus in rice . This finding provides a significant contribution to our understanding of the complex molecular mechanisms underlying hybrid sterility and microsporogenesis .

Abstract: The kidney maintains water homeostasis by modulating aquaporin 2 ( AQP2 ) on the plasma membrane of collecting duct principal cells in response to vasopressin ( VP ) . VP mediated phosphorylation of AQP2 at serine 256 is critical for this effect . However , the role of phosphorylation of other serine residues in the AQP2 C-terminus is less well understood . Here , we examined the effect of phosphorylation of S256 , S261 and S269 on AQP2 trafficking and association with recycling pathway markers . We used LLC-PK1 cells expressing AQP2 ( S-D ) or ( S-A ) phospho mutants and a 20 degrees C cold block , which allows endocytosis to continue , but prevents protein exit from the trans Golgi network ( TGN ) , inducing formation of a perinuclear AQP2 patch . AQP2-S256D persists on the plasma membrane during cold block , while wild type AQP2 , AQP2-S256A , S261A , S269A and S269D are internalized and accumulate in the patch . Development of this patch , a measure of AQP2 internalization , was most rapid with AQP2-S256A , and slowest with S261A and S269D . AQP2-S269D exhibited a biphasic internalization profile with a significant amount not internalized until 150 minutes of cold block . After rewarming to 37 degrees C , wt AQP2 , AQP2-S261A and AQP2-S269D rapidly redistributed throughout the cytoplasm within 20 minutes , whereas AQP2-S256A dissipated more slowly . Colocalization of AQP2 mutants with several key vesicular markers including clathrin , HSP70/HSC70 , EEA , GM130 and Rab11 revealed no major differences . Overall , our data provide evidence supporting the role of S256 and S269 in the maintenance of AQP2 at the cell surface and reveal the dynamics of internalization and recycling of differentially phosphorylated AQP2 in cell culture .

Abstract: Of 102 rhizoplane and endophytic bacteria isolated from rice roots and stems in California , 37% significantly ( P < or = 0 . 05 ) inhibited the growth in vitro of two pathogens , Achlya klebsiana and Pythium spinosum , causing seedling disease of rice . Four endophytic strains were highly effective against seedling disease in growth pouch assays , and these were identified as Pseudomonas fluorescens ( S3 ) , Pseudomonas tolaasii ( S20 ) , Pseudomonas veronii ( S21 ) , and Sphingomonas trueperi ( S12 ) by sequencing of amplified 16S rRNA genes . Strains S12 , S20 , and S21 contained the nitrogen fixation gene , nifD , but only S12 was able to reduce acetylene in pure culture . The four strains significantly enhanced plant growth in the absence of pathogens , as evidenced by increases in plant height and dry weight of inoculated rice seedlings relative to noninoculated rice . Three bacterial strains ( S3 , S20 , and S21 ) were evaluated in pot bioassays and reduced disease incidence by 50%-73% . Strain S3 was as effective at suppressing disease at the lowest inoculum density ( 106 CFU/mL ) as at higher density ( 10 ( 8 ) CFU/mL or undiluted suspension ) . This study indicates that selected endophytic bacterial strains have potential for control of seedling disease of rice and for plant growth promotion .

The results showed that S2 is 3512 bp long with a large open reading frame which encodes a protein of 1116 amino acids .
