Abstract: Rice blast is one of the most widespread and destructive plant diseases worldwide . Breeders have used disease resistance ( R ) genes that mediate fungal race-specific gene-for-gene resistance to manage rice blast , but the resistance is prone to breakdown due to high pathogenic variability of blast fungus . Panicle blast 1 ( Pb1 ) is a blast-resistance gene derived from the indica cultivar Modan . Pb1-mediated resistance , which is characterized by durability of resistance and adult/panicle blast resistance , has been introduced into elite varieties for commercial cultivation . We isolated the Pb1 gene by map-based cloning . It encoded a coiled-coil-nucleotide-binding-site-leucine-rich repeat ( CC-NBS-LRR ) protein . The Pb1 protein sequence differed from previously reported R-proteins , particularly in the NBS domain , in which the P-loop was apparently absent and some other motifs were degenerated . Pb1 was located within one of tandemly repeated 60-kb units , which presumably arose through local genome duplication . Pb1 transcript levels increased during the development of Pb1+ cultivars ; this expression pattern accounts for their adult/panicle resistance . Promoter : GUS analysis indicated that genome duplication played a crucial role in the generation of Pb1 by placing a promoter sequence upstream of its coding sequence , thereby conferring a Pb1-characteristic expression pattern to a transcriptionally inactive sleeping resistance gene . Abstract: The influenza A virus polymerase consists of three subunits ( PA , PB1 , and PB2 ) necessary for viral RNA synthesis . The heterotrimeric polymerase complex forms through PA interacting with PB1 and PB1 interacting with PB2 . PA has been shown to play critical roles in the assembly , catalysis , and nuclear localization of the polymerase . To probe the structure of PA , we isolated recombinant PA from insect cells . Limited proteolysis revealed that PA contained two domains connected by a 20-residue linker ( residues 257-276 ) . Far-UV circular dichroism established that the two domains folded into a mixed alpha/beta structure when separately expressed . In vitro pull-down assays showed that neither individually nor cooperatively expressed PA domains , without the linker , could assure PA-PB1 interaction . Protease treatment of PA-PB1 complex indicated that its PA subunit was significantly more stable than free PA , suggesting that the linker is protected and it constitutes an essential component of the PA-PB1 interface .

Abstract: Multiple biomarker systems have been frequently used to measure the genotoxic effects of environmental pollutants ( including heavy metals ) on living organisms . In this study , we used leaves of hydroponically grown 14 days old seedlings of rice ( Oryza sativa ) varieties ( PB1 and IR64 ) treated with 50 , 150 and 300 muM arsenite ( As ( III ) ) for 24 and 96 h duration . Reduction in seed germination , root-shoot length , chlorophyll and protein were observed with increasing As ( III ) concentration and duration in both varieties , being more in IR64 . Increase/decrease of antioxidant enzymes and stress related parameters showed much changes at higher concentration for 24 and 96 h duration in both varieties . Eleven primers were found in RAPD analysis to produce polymorphic band pattern and produced a total of 51 ( control ) , 79 ( treated ) and 42 ( control ) and 29 ( treated ) bands in PB1 and IR64 varieties , respectively . These results indicated that genomic template stability ( GTS , changes in RAPD profile ) was significantly affected at all tested As ( III ) concentration , when compared with other parameters . Differential response was observed in both varieties with PB1 being more tolerant . We concluded that DNA polymorphism detected by RAPD analysis in conjunction with other biochemical parameters could be a powerful eco-toxicological tool in bio-monitoring arsenic pollution .

Abstract: Two rice genotypes , Kalanamak 3119 ( KN3119 ) and Pusa Basmati 1 ( PB1 ) differing in their optimum nitrogen requirements ( 30 and 120 kg/ha , respectively ) were undertaken to study the expression of both high and low affinity ammonium transporter genes responsible for ammonium uptake . Exposing the roots of the seedlings of both the genotypes to increasing ( NH ( 4 ) ) ( 2 ) SO ( 4 ) concentrations revealed that all the three families of rice AMT genes are expressed , some of which get altered in a genotype and concentration specific manner . This indicates that individual ammonium transporter genes have defined contributions for ammonium uptake and plant growth . Interestingly , in response to increasing nitrogen concentrations , a root specific high affinity gene , AMT1 ; 3 , was repressed in the roots of KN3119 but not in PB1 indicating the existence of a differential ammonium sensing mechanism . This also indicates that not only AMT1 ; 3 is involved not only in ammonium uptake but may also in ammonium sensing . Further , if it can differentiate and could be used as a biomarker for nitrogen responsiveness . Expression analysis of low affinity AMT genes showed that , both AMT2 ; 1 and AMT2 ; 2 have high levels of expression in both roots and shoots and in KN3119 are induced at low ammonium concentrations . Expressions of AMT3 family genes were higher shoots than in the roots indicating that these genes are probably involved in the translocation and distribution of ammonium ions in leaves . The expression of the only high affinity AMT gene , AMT1 ; 1 , along with six low affinity AMT genes in the shoots suggests that low affinity AMTs in the shoots leaves are involved in supporting AMT1 ; 1 to carry out its activities/function efficiently .

Abstract: Two aromatic rice genotypes , Pusa Basmati 1 ( PB1 ) and Kalanamak 3119 ( KN3119 ) having 120 and 30 kg/ha optimum nitrogen requirement respectively , to produce optimal yield , were chosen to understand their differential nitrogen responsiveness . Both the genotypes grown under increasing nitrogen inputs showed differences in seed/panicle , 1 , 000 seed weight , %nitrogen in the biomass and protein content in the seeds . All these parameters in PB1 were found to be in the increasing order in contrast to KN3119 which showed declined response on increasing nitrogen dose exceeding the normal dose indicating that both the genotypes respond differentially to the nitrogen inputs . Gene expression analysis of members of ammonium transporter gene family in flag leaves during active grain filling stage revealed that all the three members of OsAMT3 family genes ( OsAMT1 ; 1-3 ) , only one member of OsAMT2 family ie , OsAMT2 ; 3 and the high affinity OsAMT1 ; 1 were differentially expressed and were affected by different doses of nitrogen . In both the genotypes , both increase and decline in seed protein contents matched with the expressions levels of OsAMT1 ; 1 , OsGS1 ; 1 and OsGS1 ; 2 in the flag leaves during grain filling stage indicating that high nitrogen nutrition in KN3119 probably causes the repression of these genes which might be important during grain filling .
