| Author: Lira-Ruan V Ruiz-Kubli M Arredondo-Peter R | Journal: Commun Integr Biol Citation: V : 4 P : 457-8 Year: 2011 Type: PubMed-not-MEDLINE | Literature: oryza Field: abstract Doc ID: pub21966570 Accession (PMID): 21966570 | Abstract: Rice ( Oryza sativa ) contains five copies of the non-symbiotic hemoglobin ( hb ) gene , namely hb1 to hb5 .
Previous analysis by RT-PCR revealed that rice hb1 expresses in roots and leaves and hb2 expresses in leaves .
However , it is not known whether or not hb1 and hb2 express in rice embryonic organs .
Here , we report the expression of hb1 and hb2 genes in rice embryonic organs using RT-PCR and specific oligos for Hb1 and Hb2 .
Our results indicate that hb1 and hb2 genes express in embryonic organs in rice growing under normal conditions .
Specifically , hb1 expresses in rice embryos and seminal roots , and hb2 expresses in embryos , coleoptiles and seminal roots .
These observations suggest that Hb1 and Hb2 coexist and function in rice embryonic organs .

Score: 3.00 | Title: Fine mapping of a major QTL for flag leaf width in rice , qFLW4 , which might be caused by alternative splicing of NAL1 .
| Author: Chen M Luo J Shao G Wei X Tang S Sheng Z Song J Hu P | Journal: Plant Cell Rep Citation: V : 31 P : 863-72 Year: 2012 Type: In-Process | Literature: oryza Field: abstract Doc ID: pub22179305 Accession (PMID): 22179305 | Abstract: Leaf width is an important agricultural trait in rice .
QTL mapping in a recombinant inbred line population derived from the cross between the javanica cultivar D50 ( narrow-leaved ) and the indica cultivar HB277 ( wide-leaved ) identified five QTLs controlling flag leaf width .
Fine mapping of the major QTL qFLW4 narrowed its location to a 74 . 8 kb interval between the SSR loci RM17483 and RM17486 , a region which also contains the gene NAL1 ( Narrow leaf 1 ) .
There was no difference in the level of NAL1 expression between cvs D50 and HB277 , but an analysis of the NAL1 transcripts showed that while most ( if not all ) of those produced in cv .
D50 were full-length , two-thirds of those in HB277 were non-functional due to either loss or gain of sequence .
The inference was that NAL1 is probably synonymous with qFLW4 , and that the functional difference between the two alleles was due to alternative splicing .
The analysis of expression of other known genes involved in the determination of leaf width provided no evidence of their having any clear functional association with qFLW4/NAL1 .

Citation: V : 115 ( 3 ) P : 1259-66 Year: 1997 Type: ARTICLE | Literature: oryza Field: abstract Doc ID: pub9390447 Accession (PMID): 9390447 | Abstract: Although nonsymbiotic hemoglobins ( Hbs ) are found in different it issues of dicots and monocots , very little is known about hb genes in monocots and the function of Hbs in nonsymbiotic it issues .
We report the cloning and analysis of two rice ( Oryza sativa L ) hb genes , hb1 and hb2 , that code for plant Hbs .
Rice hb1 and hb2 genes contain four exons and three introns , as with all of the known plant hb genes .
At least three copies of the hb gene were detected in rice DNA , and analysis of gene expression shows that hb1 and hb2 are expressed in leaves but only hb1 is expressed in roots .
A cDNA for rice Hb1 was expressed in Escherichia coli , and the recombinant Hb ( rHb1 ) shows an unusually high affinity for O2 because of a very low dissociation constant .
The absorbance spectra of the ferric and deoxyferrous rHb1 indicate that , in contrast to symbiotic Hbs , a distal ligand is coordinated to the ligand-binding site .
Mutation of the distal His demonstrates that this residue coordinates the heme Fe of ferric and deoxyferrous rHb1 and stabilizes O2 in oxy-rHb1 .
The biochemical properties of rice rHb1 suggest that this protein probably does not function to facilitate the diffusion of O2 . | Matching Sentences: [ Sen. 2, subscore: 1.00 ]: We report the cloning and analysis of two rice ( Oryza sativa L ) hb genes , hb1 and hb2 , that code for plant Hbs . | Author: Lira-Ruan V Sarath G Klucas RV Arredondo-Peter R | Journal: Citation: V : 161 ( 2 ) P : 279-287 Year: 2001 Type: ARTICLE | Literature: oryza Field: abstract Doc ID: pub11448759 Accession (PMID): 11448759 | Abstract: In rice ( Oryza sativa var .
Jackson ) at least three copies of hemoglobin ( hb ) gene exist Rice hb1 and hb2 genes are differentially expressed in roots and leaves from mature plants .
We used polyclonal antibodies raised to recombinant rice Hb1 and Western blotting to analyze the synthesis of Hbs in rice plants growing under normal or stress conditions .
Results showed that rice Hbs are synthesized in coleoptiles , seminal roots and embryos from seeds germinated for 6 days , and also in leaves and roots from plants 2-14 weeks after germination .
Analysis of Hb synthesis in stressed rice showed that : ( i ) level of Hbs was higher in etiolated than control plants , ( ii ) level of Hbs increased in roots from flooded rice , and ( iii ) level of Hbs did not change under oxidative ( H ( 2 ) O ( 2 ) ) , nitrosative ( SNP ) and hormonal ( 2 , 4-D ) stresses .
These results suggest that the effect of light withdrawal in etiolated leaves and O ( 2 ) -limiting conditions in flooded roots , but not oxidative , nitrosative and hormonal stresses , modulate the synthesis of rice Hbs . | Matching Sentences: [ Sen. 2, subscore: 1.00 ]: Jackson ) at least three copies of hemoglobin ( hb ) gene exist Rice hb1 and hb2 genes are differentially expressed in roots and leaves from mature plants .

Score: 1.00 | Title: Use of in silico ( computer ) methods to predict and analyze the tertiary structure of plant hemoglobins .
| Author: Gopalasubramaniam SK Garrocho-Villegas V Rivera GB Pastor N Arredondo-Peter R | Journal: Methods Enzymol Citation: V : 436 P : 393-410 Year: 2008 Type: MEDLINE | Literature: oryza Field: abstract Doc ID: pub18237645 Accession (PMID): 18237645 | Abstract: Amino acid sequences for more than 60 plant hemoglobins ( Hbs ) are deposited in databases , but the tertiary structure of only 4 plant Hbs have been reported ; thus , the gap between the reported sequences and structures of plant Hbs is large .
Elucidating the structure of plant Hbs is essential to fully understanding the function of these proteins in plant cells .
Determining the actual protein structure by experimental methods ( ie , by X-ray crystallography ) requires considerable protein material and is expensive ; thus , this type of work is limited to few laboratories around the world .
In silico ( computer ) methods to predict the tertiary structure of proteins from amino acid sequences have been implemented and are helping reduce the sequence-structure gap .
Thus , in silico methods are useful tools for predicting the tertiary structure of several plant Hbs from amino acid sequences deposited in databases .
In this chapter , we describe a method for predicting and analyzing the structure of a rice Hb2 from the template structure of native rice Hb1 .
This method is based on a comparative modeling method that uses programs from the SWISS-MODEL server .
