Abstract: Heading date is the one of the most important traits in rice breeding , because it defines where rice can be cultivated and influences the expression of various agronomic traits . To examine the inhibition of heading by Heading date 2 ( Hd2 ) , previously detected on the distal end of chromosome 7s long arm by quantitative trait locus ( QTL ) analysis , we developed backcross inbred lines ( BILs ) from Koshihikari , a leading Japanese cultivar , and Hayamasari , an extremely early heading cultivar . The BILs were cultivated under natural field conditions in Tsukuba Japan , and under long-day ( 14 . 5 h ) , extremely long-day ( 18 h ) , and short-day ( 10 h ) conditions . Combinations of several QTLs near Hd1 , Hd2 , Ghd7 , Hd5 , and Hd16 were detected under these four conditions . Analysis of advanced backcross progenies revealed genetic interactions between Hd2 and Hd16 and between Hd2 and Ghd7 . In the homozygous Koshihikari genetic background at Hd16 , inhibition of heading by the Koshihikari allele at Hd2 was smaller than that with the Hayamasari Hd16 allele . Similarly , in the homozygous Koshihikari genetic background at Ghd7 , the difference in heading date caused by different alleles at Hd2 was smaller than in plants homozygous for the Hayamasari Ghd7 allele . Based on these results , we conclude that Hd2 and its genetic interactions play an important role in controlling heading under long-day conditions . In addition , QTLs near Hd2 , Hd16 , and Ghd7 , which are involved in inhibition of heading under long-day conditions , function in the same pathway that controls heading date .

Abstract: In this study we have attempted to quantify the thermal and photoperiodical responses of rice ( Oryza sativa L ) flowering time QTLs jointly by a date-of-planting field experiment of a mapping population , and a phenological model analysis that separately parameterizes the two responses , based on daily temperature , daily photoperiod and flowering date . For this purpose , the three-stage Beta model , which parameterizes the sensitivity to temperature ( parameter alpha ) , the sensitivity to photoperiod ( parameter beta ) , and earliness under optimal conditions ( 10 h photoperiod at 30 degrees C ) ( parameter G ) , was applied to Nipponbare x Kasalath backcross inbred lines that were transplanted on five dates . QTLs for the beta value were detected in the four known flowering time QTL ( Hd1 , Hd2 , Hd6 and Hd8 ) regions , while QTLs for the G value were detected only in the Hd1 and Hd2 regions . This result was consistent with previous reports on near-isogenic lines ( NILs ) of Hd1 , Hd2 and Hd6 , where these loci were involved in photoperiod sensitivity , and where Hd1 and Hd2 conferred altered flowering under both 10 and 14 h photoperiods , while Hd6 action was only affected by the 14 h photoperiod . Hd8 was shown to control photoperiod sensitivity for the first time . Interestingly , Hd1 and Hd2 were associated with a QTL for the alpha value , which might support the previous hypothesis that the process of photoinduction depends on temperature . These results demonstrate that our approach can effectively quantify environmental responses of flowering time QTLs without controlled environments or NILs .

Abstract: Epigenetic phenomena have been associated with modifications of chromatin structure . These are achieved , in part , by histone post-translational modifications including acetylations and deacetylations , the later being catalyzed by histone deacetylaces ( HDACs ) . Eukaryotic HDACs are grouped into three major families , RPD3/HDA1 , SIR2 and the plant-specific HD2 . HDAC genes have been analyzed from model plants such as Arabidopsis , rice and maize and have been shown to be involved in various cellular processes including seed development , vegetative and reproductive growth and responses to abiotic and biotic stress , but reports on HDACs from other crops are limited . In this work two full-length cDNAs ( HvHDAC2-1 and HvHDAC2-2 ) encoding two members of the plant-specific HD2 family , respectively , were isolated and characterized from barley ( Hordeum vulgare ) , an agronomically important cereal crop . HvHDAC2-1 and HvHDAC2-2 were mapped on barley chromosomes 1H and 3H , respectively , which could prove useful in developing markers for marker-assisted selection in breeding programs . Expression analysis of the barley HD2 genes demonstrated that they are expressed in all it issues and seed developmental stages examined . Significant differences were observed among it issues and seed stages , and between cultivars with varying seed size , suggesting an association of these genes with seed development . Furthermore , the HD2 genes from barley were found to respond to treatments with plant stress-related hormones such as jasmonic acid ( JA ) , abscisic acid ( ABA ) and salicylic acid ( SA ) implying an association of these genes with plant resistance to biotic and abiotic stress . The expression pattern of HD2 genes suggests a possible role for these genes in the epigenetic regulation of seed development and stress response .
