Abstract: Grasses have highly specialized flowers and their outer floral organ identity remains unclear . In this study , we identified and characterized rice mutants that specifically disrupted the development of palea , one of the outer whorl floral organs . The depressed palea1 ( dp1 ) mutants show a primary defect in the main structure of palea , implying that palea is a fusion between the main structure and marginal it issues on both sides . The sterile lemma at the palea side is occasionally elongated in dp1 mutants . In addition , we found a floral organ number increase in dp1 mutants at low penetration . Both the sterile lemma elongation and the floral organ number increase phenotype are enhanced by the mutation of an independent gene SMALL DEGENERATIVE PALEA1 ( SDP1 ) , whose single mutation causes reduced palea size . E function and presumable A function floral homeotic genes were found suppressed in the dp1-2 mutant . We identified the DP1 gene by map-based cloning and found it encodes a nuclear-localized AT-hook DNA binding protein , suggesting a grass-specific role of chromatin architecture modification in flower development . The DP1 enhancer SDP1 was also positional cloned , and was found identical to the recently reported RETARDED PALEA1 ( REP1 ) gene encoding a TCP family transcription factor . We further found that SDP1/REP1 is downstreamly regulated by DP1 .

Abstract: Fructans are water-soluble fructose oligomers and polymers that are based on sucrose , and have been implicated in protecting plants against water stress . Rice ( Oryza sativa L ) is highly sensitive to chilling temperatures , and is not able to synthesize fructans . Two wheat fructan-synthesizing enzymes , sucrose : sucrose 1-fructosyltransferase , encoded by wft2 , or sucrose : fructan 6-fructosyltransferase , encoded by wft1 , were introduced into rice plants , and rice transformants that accumulate fructans were successfully obtained . The mature leaf blades of transgenic rice lines with wft2 or wft1 accumulated 16 . 2 mg g ( -1 ) FW of oligo and polysaccharides mainly composed of inulin oligomers of more than DP7 , and 3 . 7 mg g ( -1 ) FW of oligo and polysaccharides , mainly composed of phlein oligomers of more than DP15 , respectively . The transgenic rice seedlings with wft2 accumulated significantly higher concentrations of oligo and polysaccharides than non-transgenic rice seedlings , and exhibited enhanced chilling tolerance . The oligo and polysaccharide concentrations of seedlings expressing wft1 were obviously lower than those of lines expressing wft2 , and no correlation between oligo and polysaccharide concentrations and chilling tolerance was detected in wft1-expressing rice lines . The results suggest that transgenic rice lines expressing wheat-derived fructosyltransferase genes accumulated large amounts of fructans in mature leaf blades and exhibited enhanced chilling tolerance at the seedling stage . This is the first report owing that fructan accumulation enhanced tolerance to non-freezing low temperatures .

Abstract: To our knowledge the present paper shows for the first time the kinetic parameters of all the three starch branching enzyme ( BE ) isozymes , BEI , BEIIa and BEIIb , from rice with both amylopectin and synthetic amylose as glucan substrate . The activities of these BE isozymes with a linear glucan amylose decreased with a decrease in the molar size of amylose , and no activities of BEIIa and BEIIb were found when the degree of polymerization ( DP ) of amylose was lower than at least 80 , whereas BEI had an activity with amylose of a DP higher than approximately 50 . Detailed analyses of debranched products from BE reactions revealed the distinct chain length preferences of the individual BE isozymes .
