Abstract: The it issue-specific expression of transgenes is essential in plant breeding programmes to avoid the fitness costs caused by constitutive expression of a target gene . However , knowledge on the molecular mechanisms of it issue-specific gene expression and practicable it issue-specific promoters is limited . In this study , we identified the cis-acting elements of a it issue-specific promoter from rice , P ( D54O ) , and tested the application of original and modified P ( D54O ) and its cis-elements in the regulation of gene expression . P ( D54O ) is a green it issue-specific promoter . Five novel it issue-specific cis-elements ( LPSE1 , LPSE2 , LPSRE1 , LPSRE2 , PSE1 ) were characterized from P ( D54O ) . LPSE1 activated gene expression in leaf and young panicle . LPSRE2 suppressed gene expression in leaf , root , young panicle and stem , and PSE1 suppressed gene expression in young panicle and stem . LPSRE1 and LPSE2 had dual roles in the regulation of it issue-specific gene expression ; both functioned as activators in leaf , but LPSRE1 acted as a repressor in stem and LPSE2 as a repressor in young panicle and root . Transgenic rice plants carrying cry1Ac encoding Bacillus thuringiensis endotoxin , regulated by P ( D54O ) , were resistant to leaf-folders , with no Cry1Ac protein found in endosperm or embryo . A reporter gene regulated by a series of truncated P ( D54O ) showed various it issue-specific expression patterns . Different fragments of P ( D54O ) fused with the constitutive cauliflower mosaic virus 35S promoter suppressed 35S-regulated gene expression in various it issues . P ( D54O ) , truncated P ( D54O ) and the it issue-specific cis-elements provide useful tools for the regulation of it issue-specific gene expression in rice breeding programmes .

Abstract: Varietal mutants can be useful for developing durable resistance , understanding categories of resistance , and identifying candidate genes involved in defense responses . We used mutants of rice IR64 to isolate new sources of resistance to the planthopper Nilaparvata lugens ( Stal ) ( Hemiptera : Delphacidae ) . We compared two mutants that showed a gain and loss of resistance to N lugens , to determine the categories of resistance to this pest Under choice tests , female planthoppers avoided settling and laid fewer eggs on the resistant mutant D518 than on the susceptible mutant D1131 , susceptible check TN1 , and wild-type IR64 , indicating that antixenosis was the resistance category . Similarly , under no-choice conditions , planthoppers laid 29% fewer eggs in D518 than in IR64 , but they oviposited more in D1131 and TN1 . Honeydew excretion was greater on D1131 seedlings but slightly lower on D518 than on IR64 . Nymphal survival and adult female weight did not differ among rice cultivars . D518 showed higher tolerance of N lugens infestations than IR64 . Genetic analysis of the F1 , F2 , and F3 populations derived from D518 x IR64 revealed that resistance in D518 is dominant and controlled by a single gene . Despite the variation in resistance to N lugens , both mutants and IR64 performed similarly in the field . The mutant D518 is a new source of durable resistance to N lugens , mainly due to enhanced antixenosis to female hoppers for settling and oviposition .

Abstract: Rice internodes are vital for supporting high-yield panicles , which are controlled by various factors such as cell division , cell elongation and cell wall biosynthesis . Therefore , formation and regulation of the internode cell-producing intercalary meristem ( IM ) are important for determining the shape of internodes . To understand the regulation of internode development , we analysed a rice dwarf mutant , dwarf 50 ( d50 ) . Previously , we reported that parenchyma cells in the elongated internodes of d50 ectopically deposit cell wall phenolics . In this study , we revealed that D50 encodes putative inositol polyphosphate 5-phosphatase ( 5PTase ) , which may be involved in phosphoinositide signalling required for many essential cellular functions , such as cytoskeleton organization , endocytosis and vesicular trafficking in eukaryotes . Analysis of the rice genome revealed 20 putative 5PTases including D50 . The d50 mutation induced abnormally oriented cell division , irregular deposition of cell wall pectins and thick actin bundles in the parenchyma cells of the IM , resulting in abnormally organized cell files of the internode parenchyma and dwarf phenotype . Our results suggest that the putative 5PTase , encoded by D50 , is essential for IM formation , including the direction of cell division , deposition of cell wall pectins and control of actin organization .

Abstract: To characterize the genetic basis of drug resistance in Mycobacterium tuberculosis in Latvia , mutations involved in rifampin ( rpoB gene ) and isoniazid ( katG gene ) resistance in DNA from 19 drug-susceptible and 51 multidrug-resistant M tuberculosis complex isolates were analyzed . Nucleotide substitution at codon 315 ( AGC-->ACC ) of the katG gene was found in 48 of 51 multidrug-resistant samples by sequencing .
