Abstract: A novel gene for alternative oxidase ( AOX ) was isolated from rice ( Oryza sativa L ) and characterized . The deduced amino acid sequence of the novel AOX gene contains features that are conserved among other AOXs . This AOX gene was designated AOX1c based on a phylogenetic analysis of the AOX genes . Northern hybridization analyses revealed that AOX1c and AOX1a/AOX1b transcripts accumulated differently in various rice organs and rice seedlings under low temperature conditions . AOX1c mRNA was mainly present in young leaves under constant light , mature leaves and panicles after heading , but it was not detected in young etiolated leaves and young roots of seedlings or young panicles . On the other hand , the mRNAs of the rice AOX1a and AOX1b genes were mainly present in young roots and mature leaves . Under low temperature conditions , the steady-state mRNA levels of the rice AOX1a and AOX1b genes clearly increased with time but the rice AOX1c gene was apparently not responsive to low temperature . The rice AOX gene family and differences in their regulation are discussed .

Abstract: A new type of transposon , named Anaconda ( Anac ) has been found in rice ( Oryza sativa ) . In this paper , we demonstrate that Anaconda elements have diversified by acquisition of host cellular genes , amplification of the elements , and substitution and deletion of short segments . We identified four Anaconda elements in studies of rice alternative oxidase ( AOX ) genes , and subsequently isolated an additional 23 elements based on the identity of their terminal inverted repeats ( TIRs ) . The Anaconda elements have long TIRs ( 114-458 bp ) . They also have direct repeats of 9 or 10 bp in their flanking regions that are thought to have been generated upon transposition . These structural features reveal that the Anaconda elements belong to the Mu superfamily . The most prominent feature of the Anaconda elements is the high frequency with which they have acquired host cellular genes . Of the 27 elements found here , 19 appear to have sequences presumably derived from rice genes , for example , the genes for AOX1c ( four elements ) , cytochrome P450 ( five elements ) , L : -asparaginase ( five elements ) , and PCF8 ( two elements ) . Four elements , AnacA1-A4 , have both the AOX1c and P450 genes . One element , AnacB14 , involves a gene similar to mudrA of maize MuDR . Database analyses revealed that the loci of 26 of the 27 Anaconda elements in the subspecies japonica are the same as those in the subspecies indica . This suggests that these elements were incorporated before the divergence of these two subspecies .

Abstract: Under constant light , increases in V ( t ) ( total respiration rate ) , V ( alt ) ( capacity of alternative pathway ) , and V ( alt ) /V ( t ) ration values ( Figs1-3 ) in fully etiolated rice seedlings leaves were founded . Northern hybridization with special probes for rice AOX1a , AOX1b and AOX1c revealed that the AOX1c mRNA was mainly present under constant light ( Fig4 ) . SHAM ( salicylhydroxamic acid ) decreased the relative oxygen evolution rate and chlorophyll content during the rapid greening of the etiolated rice seedlings ( Figs6 , 7 ) . We found that illumination caused bigger increase in V ( alt ) in fully etiolated rice seedlings than in any partially etiolated rice seedlings ( Fig8A ) , while the soluble sugar contents did not show any significant differences ( Fig8B ) . This revealed the connection between greening process and cyanide-resistant respiration in rice seedlings . We infer that one of the possible functions of the cyanide-resistant respiration is to maintain a high rate of respiratory electron transport for the essential biosynthesis processes at the early stage of chloroplast development . Cyanide-resistant respiration may provide a regulatory mechanism for the balance between the requirements of the energy and substances .

Abstract: Mitochondrial alternative oxidase ( AOX ) is the terminal oxidase responsible for cyanide-insensitive and salicylhydroxamic acid-sensitive respiration . We have isolated two non-homoeologous genes ( Waox1a and Waox1c ) encoding AOX proteins from common wheat ( Triticum aestivum L ) . These two genes were orthologous to rice AOX1a and AOX1c , and their exon/intron structure was conserved , as it is in most other plant AOX genes . Southern blot analysis indicated that both Waox1a and Waox1c were located in at least three homoeologous loci and that additional AOX genes with lower homology were present in the genome of common wheat . The Waox1a and Waox1c loci were respectively assigned to the homoeologous group 2 and 6 chromosomes . Abstract: Alternative respiratory pathway was investigated in rice seedlings grown under total darkness , light/dark cycle , or continuous light . The capacity of the alternative pathway was relatively higher in leaves that had longer light exposure . An analysis of rice AOX1 multigene family revealed that AOX1c , but not AOX1a and AOX1b , had a light-independent expression . The alternative oxidase ( AOX ) inhibitor , salicylhydroxamic acid ( SHAM , 1mM ) , inhibited nearly 68% of the capacity of the alternative pathway in leaves grown under different light conditions . The plants grown under different light periods were treated with SHAM and then were exposed to illumination for 4h . The transition from dark to 4h of light stimulated the capacity of alternative pathway in etiolated rice seedlings and in those grown under light/dark cycle , whereas the capacity of the alternative pathway was constant in seedlings grown under continuous light with additional 4h of illumination . Etiolated leaves did not show any CO ( 2 ) fixation after 4h of illumination , and the increase in chlorophyll content was delayed by the SHAM pretreatment . When seedlings grown under light/dark cycle were moved from dark and exposed to 4h of light , increases in chlorophyll content and CO ( 2 ) fixation rate were reduced by SHAM . Although these parameters were stable in plants grown under continuous light , SHAM decreased CO ( 2 ) fixation rate but not the chlorophyll content . These results indicate that the role and regulation of AOX in light are determined by the developmental stage of plant photosynthetic apparatus .
