Abstract: In rice ( Oryza sativa var . Jackson ) at least three copies of hemoglobin ( hb ) gene exist Rice hb1 and hb2 genes are differentially expressed in roots and leaves from mature plants . We used polyclonal antibodies raised to recombinant rice Hb1 and Western blotting to analyze the synthesis of Hbs in rice plants growing under normal or stress conditions . Results showed that rice Hbs are synthesized in coleoptiles , seminal roots and embryos from seeds germinated for 6 days , and also in leaves and roots from plants 2-14 weeks after germination . Analysis of Hb synthesis in stressed rice showed that : ( i ) level of Hbs was higher in etiolated than control plants , ( ii ) level of Hbs increased in roots from flooded rice , and ( iii ) level of Hbs did not change under oxidative ( H ( 2 ) O ( 2 ) ) , nitrosative ( SNP ) and hormonal ( 2 , 4-D ) stresses . These results suggest that the effect of light withdrawal in etiolated leaves and O ( 2 ) -limiting conditions in flooded roots , but not oxidative , nitrosative and hormonal stresses , modulate the synthesis of rice Hbs .

Abstract: Amino acid sequences for more than 60 plant hemoglobins ( Hbs ) are deposited in databases , but the tertiary structure of only 4 plant Hbs have been reported ; thus , the gap between the reported sequences and structures of plant Hbs is large . Elucidating the structure of plant Hbs is essential to fully understanding the function of these proteins in plant cells . Determining the actual protein structure by experimental methods ( ie , by X-ray crystallography ) requires considerable protein material and is expensive ; thus , this type of work is limited to few laboratories around the world . In silico ( computer ) methods to predict the tertiary structure of proteins from amino acid sequences have been implemented and are helping reduce the sequence-structure gap . Thus , in silico methods are useful tools for predicting the tertiary structure of several plant Hbs from amino acid sequences deposited in databases . In this chapter , we describe a method for predicting and analyzing the structure of a rice Hb2 from the template structure of native rice Hb1 . This method is based on a comparative modeling method that uses programs from the SWISS-MODEL server .

Abstract: Leaf width is an important agricultural trait in rice . QTL mapping in a recombinant inbred line population derived from the cross between the javanica cultivar D50 ( narrow-leaved ) and the indica cultivar HB277 ( wide-leaved ) identified five QTLs controlling flag leaf width . Fine mapping of the major QTL qFLW4 narrowed its location to a 74 . 8 kb interval between the SSR loci RM17483 and RM17486 , a region which also contains the gene NAL1 ( Narrow leaf 1 ) . There was no difference in the level of NAL1 expression between cvs D50 and HB277 , but an analysis of the NAL1 transcripts showed that while most ( if not all ) of those produced in cv . D50 were full-length , two-thirds of those in HB277 were non-functional due to either loss or gain of sequence . The inference was that NAL1 is probably synonymous with qFLW4 , and that the functional difference between the two alleles was due to alternative splicing . The analysis of expression of other known genes involved in the determination of leaf width provided no evidence of their having any clear functional association with qFLW4/NAL1 .

Abstract: Single chain variable fragment antibody ( scFv ) is capable of binding its target antigens and is one of the most popular recombinant antibodies format for many applications . In this study , a large human synthetic phage displayed library ( Tomlinson J ) was employed to generate scFvs against Cry1B toxin by affinity panning . After four rounds of panning , six monoclonal phage particles capable of binding with the Cry1B were isolated , sequenced and characterized by Enzyme-Linked Immunosorbent Assay ( ELISA ) . Two of the identified novel anti-Cry1B scFvs , namely H9 and B12 , were expressed in Escherichia coli HB2151 and purified by Ni metal ion affinity chromatography . Sodium dodecyl sulfate polyacrylamine gel electrophoresis ( SDS-PAGE ) indicated that the relative molecular mass of scFv was estimated at 30 kDa . The purified scFv-H9 was used to develop an indirect competitive enzyme-linked immunosorbent assay ( ic-ELISA ) for Cry1B toxin . The linear range of detection for standards in this ic-ELISA was approximately 0 . 19-1 . 1 mug mL ( - ) ( 1 ) and 50% inhibition of control ( IC ( 5 ) ( 0 ) ) was 0 . 84 mug mL ( - ) ( 1 ) for Cry1B . The affinity of scfv-H9 was ( 1 . 95+/-0 . 12 ) x 10 ( 7 ) M ( - ) ( 1 ) and showed cross-reactivity with Cry1Ab toxin and Cry1Ac toxin ( 8 . 53% and 7 . 58% , respectively ) , higher cross-reactivity ( 12 . 8% ) with Cry1C toxin . The average recoveries of Cry1B toxin from spiked leaf and rice samples were in the range 89 . 5-96 . 4% , and 88 . 5-95 . 6% , respectively , with a coefficient of variation ( CV ) less than 6 . 0% . These results showed promising applications of scfv-H9 for detecting Cry1B toxin in agricultural and environmental samples .
