Abstract: Rice ragged stunt oryzavirus ( RRSV ) is a member of the genus oryzavirus within the family Reoviridae . Its genome consists of ten segments of dsRNA . The functions of all products encoded by these viral genome segments , except one encoded by S9 , have not yet been elucidated . In the present study , the ORF of S 8 of RRSV-Philippines isolate was sequenced and expressed in E coli . The 67 kD product of S8 could be self-cleaved into two fragments with molecular weights of 43 kD and 26 kD . Western blotting indicated that both 67 kD and 43 kD products were major structural proteins of the virus . It was also found that the 67 kD protein could self aggregate into aggregates with higher sedimentation rate in sucrose gradients during centrifugation . Moreover , the self-aggregation process could be accelerated by the complex of S6 product and genome dsRNAs of RRSV . These results suggest that the S8 products , 67 kD or 43 kD , may be the structural components of the viral inner-capsid .

Abstract: This is the first sequence-based characterization of Mal de R Cuarto virus ( MRCV ) , currently classified as a variant of Maize rough dwarf virus ( MRDV ) and exclusively found in South America . We sequenced and analyzed genome segments S4 and S8 . MRCV S4 coded for a putative 131 . 67 kDa protein while MRCV S8 coded for a putative 68 . 26 kDa protein containing an ATP/GTP-binding motif . The 5 and 3 ends of MRCV segments , were 5AAGUUUUU3 and 5CAGCUnnnGUC3 , respectively . Prediction of secondary structure of both segments coding strands showed that terminal regions were able to form structures that are proposed to be replication and packaging signals . MRCV S4 showed identity to members of Fijivirus as well as to two other genera of the Reoviridae family . MRCV S8 revealed identity with Rice black streaked dwarf virus ( RBSDV ) S8 , MRDV S7 , Oat sterile dwarf virus ( OSDV ) S9 and Nilaparvata lugens reovirus ( NLRV ) S7 . While MRDV and RBSDV segments are highly homologous between each other , MRCV identity levels with them was considerably lower . We discussed the evolutionary relationships of MRCV to other Reoviridae , and based on phylogenetic analysis we proposed that although MRCV is related to MRDV , it could be regarded as a new species of the Fijivirus genus .

Abstract: The double strand RNA of maize rough dwarf virus ( MRDV ) was prepared from the maize samples showing symptoms which was from the Luanchen county of Heibei province of China . The primers were designed according to the known sequence of MRDV , the cDNA sequence of the seventh segment of MRDV was obtained by RT-PCR , the S7 sequence was analyzed by computer after sequencing . Abstract: The Arabidopsis thaliana genome has over 550 protease sequences representing all five catalytic types : serine , cysteine , aspartic acid , metallo and threonine ( MEROPS peptidase database , http : //merops . sanger . ac . uk/ ) , which probably reflect a wide variety of as yet unidentified functions performed by plant proteases . Recent indications that the 26S proteasome , a T1 family-threonine protease , is a regulator of light and hormone responsive signal transduction highlight the potential of proteases to participate in many aspects of plant growth and development . Recent discoveries that proteases are required for stomatal distribution , embryo development and disease resistance point to wider roles for four additional multigene families that include some of the most frequently studied ( yet poorly understood ) plant proteases : the subtilisin-like , serine proteases ( family S8 ) , the papain-like , cysteine proteases ( family C1A ) , the pepsin-like , aspartic proteases ( family A1 ) and the plant matrixin , metalloproteases ( family M10A ) . In this report , 54 subtilisin-like , 30 papain-like and 59 pepsin-like proteases from Arabidopsis , are compared with S8 , C1A and A1 proteases known from other plant species at the functional , phylogenetic and gene structure levels . Examples of structural conservation between S8 , C1A and A1 genes from rice , barley , tomato and soybean and those from Arabidopsis are noted , indicating that some common , essential plant protease roles were established before the divergence of monocots and eudicots . Numerous examples of tandem duplications of protease genes and evidence for a variety of restricted expression patterns suggest that a high degree of specialization exists among proteases within each family . We propose that comprehensive analysis of the functions of these genes in Arabidopsis will firmly establish serine , cysteine and aspartic proteases as regulators and effectors of a wide range of plant processes .

Abstract: Rice dwarf virus ( RDV ) , a member of the family Reoviridae , has a 12-segmented dsRNA genome . Seven segments , designated S1 , S2 , S3 , S5 , S7 , S8 , and S9 , encode structural proteins , while the remainder encode nonstructural proteins . One of the nonstructural proteins , Pns4 , which is encoded by S4 , was characterized . Pns4 was a phosphorylatable substrate in a phosphorylation assay in vivo ; it associated with large cytoplasmic fibrils and formed novel minitubules in infected cultured cells of its leafhopper insect vector , as revealed by immunofluorescence and immunoelectron microscopy . Early in infection , Pns4 was detected at the periphery of the viroplasm , and it was then observed on amorphous or fibrillar inclusions , which were identified as bundles of minitubules , at later stages of infection . Since viroplasms are believed to be the site of RDV replication , the intracellular location of Pns4 suggests that this protein might be involved in the process of assembly of the RDV virion .

Abstract: Although pronounced heterosis in inter-subspecific hybrids was known in rice for a long time , its utilization for hybrid rice breeding has been limited due to their hybrid sterility ( HS ) . For the last two decades , however , a few inter-subspecific hybrids have been developed by incorporating wide-compatibility genes ( WCG ) that resolve HS , into parental lines of these inter-subspecific hybrids . For effective use of WCG , it is necessary to find convenient markers linked to WCG of practical importance . In this paper , initially a set of simple sequence repeat ( SSR ) markers in the vicinity of known WCG loci identified based on comparative linkage maps have been surveyed in a population derived from the three-way cross IR36/Dular//Akihikari , where a known donor of WCG Dular was crossed to a representative indica and japonica cultivar . Of the five parental polymorphic markers , RM253 and RM276 were found to be closely linked to the WCG locus S5 at a distance of 3 . 0 and 2 . 8 cM , respectively . Later , loci for HS were examined in three F ( 2 ) populations derived from inter-subspecific crosses , with same set of SSR markers . The locus S8 was confirmed to have major influence on HS in the F ( 2 ) population derived from CHMRF-1/Taichung65 since two SSR markers in its vicinity , RM412 and RM141 , co-segregated with HS at a map distance of 7 . 6 and 4 . 8 cM , respectively . In the F ( 2 ) population derived from the cross BPT5204/Taipei309 , three SSR markers in the vicinity of S5 , RM50 , RM276 and RM136 co-segregated with HS at a map distance of 4 . 2 , 3 . 2 and 7 . 8 cM , respectively . In the third F ( 2 ) population derived from Swarna/Taipei309 , the SSR markers in the vicinity of S5 , RM225 , RM253 , RM50 , RM276 and RM136 were identified to co-segregate with HS at a map distance of 3 . 2 , 2 . 6 , 3 . 4 , 2 . 6 and 6 . 6 cM , respectively . These results indicated a clear picture of WCG in Dular as well as the predominant role of HS alleles at S5 locus . The identified SSR markers are expected to be used for incorporation of WCG into parental lines in hybrid rice breeding to solve HS in inter-subspecific hybrids .

Abstract: OBJECTIVE : To estimate the likelihood of horizontal gene transfer from transgenic rice to bacteria of the food chain and human gut , the stability of s86 transgenic rice hpt gene in different food matrices and under varying food-processing conditions was studied . METHODS : Degradation of DNA was monitored by fragment -multiplex polymerase chain reaction . Integrity of hpt gene in various food samples was tested . RESULTS : A PCR system for the hpt gene of genetically modified rice has been established to detect fragments ranging between 236bp and 910bp . Detection of hpt and rbcl gene fragments was carried out in various food-processed samples by this PCR system . The data showed that the fragments up to 500 bp were detected in rice and congee , while the fragment length more than 236bp was not detected in crispy rice and popcorn-like rice . CONCLUSION : These results suggested that there are significant differences in DNA degradation by different food-processing methods . The likelihood of the large hpt gene fragments transfer from transgenic rice processed food to bacteria is reduced by food process .

Abstract: Tobacco leaf enation virus ( TLEV ) is a putative member of the genus Phytoreovirus within the family Reoviridae . Previous western blot analysis of structural viral proteins ( apparent molecular weights of 93 kDa ; 58 kDa ; 48 kDa ; 39 kDa and 36 kDa ) associated with TLEV , isolated from infected tobacco in South Africa , suggested that these proteins may correspond to structural Wound tumor virus ( WTV ) proteins . To further establish the nature of this novel virus disease phenotype in tobacco , molecular characterization of six dsRNA components was undertaken . Full-length cDNA clones were obtained by an optimized modified single-primer amplification sequence-independent dsRNA cloning method . Results of this study revealed the conserved terminal sequence : 5GG ( U/C ) . . . UGAU 3 of segments S6-S12 , while adjacent to these conserved terminal sequences are imperfect inverted repeats ( 7-15 bp in length ) , both features being common to reoviruses . The complete nucleotide sequences of segments S5 ( 2 , 610 bp ) , S7 ( 1 , 740 bp ) , S8 ( 1 , 439 bp ) , S10 ( 1 , 252 bp ) , S11 ( 1 , 187 bp ) and S12 ( 836 bp ) were determined . Comparison of full-length nucleotide sequences with corresponding segments of other phytoreoviruses , Rice gall dwarf virus ( RGDV ) , Rice dwarf virus ( RDV ) and WTV has shown nucleotide and predicted amino acid identities within the range of 30-60% . TLEV consistently shows a higher identity to WTV than to other phytoreovirus species where sequence data is available .
