Abstract: Varietal mutants can be useful for developing durable resistance , understanding categories of resistance , and identifying candidate genes involved in defense responses . We used mutants of rice IR64 to isolate new sources of resistance to the planthopper Nilaparvata lugens ( Stal ) ( Hemiptera : Delphacidae ) . We compared two mutants that showed a gain and loss of resistance to N lugens , to determine the categories of resistance to this pest Under choice tests , female planthoppers avoided settling and laid fewer eggs on the resistant mutant D518 than on the susceptible mutant D1131 , susceptible check TN1 , and wild-type IR64 , indicating that antixenosis was the resistance category . Similarly , under no-choice conditions , planthoppers laid 29% fewer eggs in D518 than in IR64 , but they oviposited more in D1131 and TN1 . Honeydew excretion was greater on D1131 seedlings but slightly lower on D518 than on IR64 . Nymphal survival and adult female weight did not differ among rice cultivars . D518 showed higher tolerance of N lugens infestations than IR64 . Genetic analysis of the F1 , F2 , and F3 populations derived from D518 x IR64 revealed that resistance in D518 is dominant and controlled by a single gene . Despite the variation in resistance to N lugens , both mutants and IR64 performed similarly in the field . The mutant D518 is a new source of durable resistance to N lugens , mainly due to enhanced antixenosis to female hoppers for settling and oviposition .

Abstract: To characterize the genetic basis of drug resistance in Mycobacterium tuberculosis in Latvia , mutations involved in rifampin ( rpoB gene ) and isoniazid ( katG gene ) resistance in DNA from 19 drug-susceptible and 51 multidrug-resistant M tuberculosis complex isolates were analyzed . The most frequent rpoB gene mutations found by the Line Probe assay were the S531L ( 14 of 34 isolates ) , D516V ( 7 of 34 ) , H526D ( 4 of 34 ) , and D516Y plus P535S ( 4 of 34 ) mutations . Direct sequencing of seven isolates with unclear results from Line Probe assay showed the presence of the L533P mutation and the Q510H plus H526Y ( 1 of 34 ) and D516V plus P535S ( 4 of 34 ) double mutations , neither of which has been described previously . Single-strand conformation polymorphism analysis showed strand mobility differences between the rifampin-susceptible and -resistant samples for the D516V , H526D , and D516Y plus P535S mutations but not for the S531L mutation . Nucleotide substitution at codon 315 ( AGC-->ACC ) of the katG gene was found in 48 of 51 multidrug-resistant samples by sequencing . Furthermore , katG gene restriction fragment length polymorphism analysis with endonuclease AciI confirmed the nucleotide change in codon 315 .

Abstract: The inheritance of plant height was investigated in a ten-parent diallel cross of diverse rice cultivars . Parents included two tall japonica lines and eight semidwarf lines . Data from parent , F ( 1 ) , F ( 2 ) , and F ( 3 ) generations indicated that the majority of height variation among the ten parents could be accounted for by three major genes with additive loci effects . D51 , 72/2234-11 , and G33 ( derived from the known major-gene indica semidwarf Dee-geo-woo-gen ) all were found to possess an allelic , partially recessive semidwarfing gene ( sd ( 1 ) ) . Additional semidwarfing genes were detected in D66 ( sd ( 2 ) , fully recessive ) and in CI 9858 ( sd ( 3 ) , partially to fully recessive ) . Relative magnitudes of additive effects were sd ( 1 ) > sd ( 2 ) >/= sd ( 3 ) . Hokuriki 76 , Tedoriwase , and IV 29-4 were found to be dwarfed by a multiple-gene system . Hayman-Jinks diallel cross analysis on parent and F ( 1 ) information ( 1974 and 1975 ) and on parent and F ( 2 ) information demonstrated the presence of significant additive and dominance variation , but epistasis was not detected .
