Abstract: Rice ragged stunt oryzavirus ( RRSV ) is a member of the genus oryzavirus within the family Reoviridae . Its genome consists of ten segments of dsRNA . The functions of all products encoded by these viral genome segments , except one encoded by S9 , have not yet been elucidated . In the present study , the ORF of S 8 of RRSV-Philippines isolate was sequenced and expressed in E coli . The 67 kD product of S8 could be self-cleaved into two fragments with molecular weights of 43 kD and 26 kD . Western blotting indicated that both 67 kD and 43 kD products were major structural proteins of the virus . It was also found that the 67 kD protein could self aggregate into aggregates with higher sedimentation rate in sucrose gradients during centrifugation . Moreover , the self-aggregation process could be accelerated by the complex of S6 product and genome dsRNAs of RRSV . These results suggest that the S8 products , 67 kD or 43 kD , may be the structural components of the viral inner-capsid .

Title: Sequence analysis of genome segments S4 and S8 of Mal de R Cuarto virus ( MRCV ) : evidence that the virus should be a separate Fijivirus species .

Abstract: This is the first sequence-based characterization of Mal de R Cuarto virus ( MRCV ) , currently classified as a variant of Maize rough dwarf virus ( MRDV ) and exclusively found in South America . We sequenced and analyzed genome segments S4 and S8 . MRCV S4 coded for a putative 131 . 67 kDa protein while MRCV S8 coded for a putative 68 . 26 kDa protein containing an ATP/GTP-binding motif . The 5 and 3 ends of MRCV segments , were 5AAGUUUUU3 and 5CAGCUnnnGUC3 , respectively . Prediction of secondary structure of both segments coding strands showed that terminal regions were able to form structures that are proposed to be replication and packaging signals . MRCV S4 showed identity to members of Fijivirus as well as to two other genera of the Reoviridae family . MRCV S8 revealed identity with Rice black streaked dwarf virus ( RBSDV ) S8 , MRDV S7 , Oat sterile dwarf virus ( OSDV ) S9 and Nilaparvata lugens reovirus ( NLRV ) S7 . While MRDV and RBSDV segments are highly homologous between each other , MRCV identity levels with them was considerably lower . We discussed the evolutionary relationships of MRCV to other Reoviridae , and based on phylogenetic analysis we proposed that although MRCV is related to MRDV , it could be regarded as a new species of the Fijivirus genus .

Abstract: The Arabidopsis thaliana genome has over 550 protease sequences representing all five catalytic types : serine , cysteine , aspartic acid , metallo and threonine ( MEROPS peptidase database , http : //merops . sanger . ac . uk/ ) , which probably reflect a wide variety of as yet unidentified functions performed by plant proteases . Recent indications that the 26S proteasome , a T1 family-threonine protease , is a regulator of light and hormone responsive signal transduction highlight the potential of proteases to participate in many aspects of plant growth and development . Recent discoveries that proteases are required for stomatal distribution , embryo development and disease resistance point to wider roles for four additional multigene families that include some of the most frequently studied ( yet poorly understood ) plant proteases : the subtilisin-like , serine proteases ( family S8 ) , the papain-like , cysteine proteases ( family C1A ) , the pepsin-like , aspartic proteases ( family A1 ) and the plant matrixin , metalloproteases ( family M10A ) . In this report , 54 subtilisin-like , 30 papain-like and 59 pepsin-like proteases from Arabidopsis , are compared with S8 , C1A and A1 proteases known from other plant species at the functional , phylogenetic and gene structure levels . Examples of structural conservation between S8 , C1A and A1 genes from rice , barley , tomato and soybean and those from Arabidopsis are noted , indicating that some common , essential plant protease roles were established before the divergence of monocots and eudicots . Numerous examples of tandem duplications of protease genes and evidence for a variety of restricted expression patterns suggest that a high degree of specialization exists among proteases within each family . We propose that comprehensive analysis of the functions of these genes in Arabidopsis will firmly establish serine , cysteine and aspartic proteases as regulators and effectors of a wide range of plant processes .

Abstract: The complete sequences of Fiji disease virus ( FDV ) genome segments 5 ( S5 ) , S6 , S8 and S10 were obtained and comprised 3150 nt , 2831 nt , 1959 nt and 1819 nt , respectively . Each segment contained a single ORF which encoded putative proteins of 115 kDa , 97 kDa , 69 kDa and 63 . 0 kDa , respectively . The putative amino acid sequences encoded by S5 and S6 contained putative leucine zipper motifs while FDV S5 and S8 each contained an ATP-GTP-binding motif . At the amino acid level , FDV S5 , S6 , S8 and S10 showed most similarity to the corresponding segments of Rice black-streaked dwarf virus . Based on sequence similarities , it is predicted that FDV S8 encodes a minor core protein , while FDV S10 encodes an outer capsid protein . Abstract: Rice dwarf virus ( RDV ) , a member of the family Reoviridae , has a 12-segmented dsRNA genome . Seven segments , designated S1 , S2 , S3 , S5 , S7 , S8 , and S9 , encode structural proteins , while the remainder encode nonstructural proteins . One of the nonstructural proteins , Pns4 , which is encoded by S4 , was characterized . Pns4 was a phosphorylatable substrate in a phosphorylation assay in vivo ; it associated with large cytoplasmic fibrils and formed novel minitubules in infected cultured cells of its leafhopper insect vector , as revealed by immunofluorescence and immunoelectron microscopy . Early in infection , Pns4 was detected at the periphery of the viroplasm , and it was then observed on amorphous or fibrillar inclusions , which were identified as bundles of minitubules , at later stages of infection . Since viroplasms are believed to be the site of RDV replication , the intracellular location of Pns4 suggests that this protein might be involved in the process of assembly of the RDV virion .

Abstract: Although pronounced heterosis in inter-subspecific hybrids was known in rice for a long time , its utilization for hybrid rice breeding has been limited due to their hybrid sterility ( HS ) . For the last two decades , however , a few inter-subspecific hybrids have been developed by incorporating wide-compatibility genes ( WCG ) that resolve HS , into parental lines of these inter-subspecific hybrids . For effective use of WCG , it is necessary to find convenient markers linked to WCG of practical importance . In this paper , initially a set of simple sequence repeat ( SSR ) markers in the vicinity of known WCG loci identified based on comparative linkage maps have been surveyed in a population derived from the three-way cross IR36/Dular//Akihikari , where a known donor of WCG Dular was crossed to a representative indica and japonica cultivar . Of the five parental polymorphic markers , RM253 and RM276 were found to be closely linked to the WCG locus S5 at a distance of 3 . 0 and 2 . 8 cM , respectively . Later , loci for HS were examined in three F ( 2 ) populations derived from inter-subspecific crosses , with same set of SSR markers . The locus S8 was confirmed to have major influence on HS in the F ( 2 ) population derived from CHMRF-1/Taichung65 since two SSR markers in its vicinity , RM412 and RM141 , co-segregated with HS at a map distance of 7 . 6 and 4 . 8 cM , respectively . In the F ( 2 ) population derived from the cross BPT5204/Taipei309 , three SSR markers in the vicinity of S5 , RM50 , RM276 and RM136 co-segregated with HS at a map distance of 4 . 2 , 3 . 2 and 7 . 8 cM , respectively . In the third F ( 2 ) population derived from Swarna/Taipei309 , the SSR markers in the vicinity of S5 , RM225 , RM253 , RM50 , RM276 and RM136 were identified to co-segregate with HS at a map distance of 3 . 2 , 2 . 6 , 3 . 4 , 2 . 6 and 6 . 6 cM , respectively . These results indicated a clear picture of WCG in Dular as well as the predominant role of HS alleles at S5 locus . The identified SSR markers are expected to be used for incorporation of WCG into parental lines in hybrid rice breeding to solve HS in inter-subspecific hybrids .

Abstract: OBJECTIVE : To estimate the likelihood of horizontal gene transfer from transgenic rice to bacteria of the food chain and human gut , the stability of s86 transgenic rice hpt gene in different food matrices and under varying food-processing conditions was studied . METHODS : Degradation of DNA was monitored by fragment -multiplex polymerase chain reaction . Integrity of hpt gene in various food samples was tested . RESULTS : A PCR system for the hpt gene of genetically modified rice has been established to detect fragments ranging between 236bp and 910bp . Detection of hpt and rbcl gene fragments was carried out in various food-processed samples by this PCR system . The data showed that the fragments up to 500 bp were detected in rice and congee , while the fragment length more than 236bp was not detected in crispy rice and popcorn-like rice . CONCLUSION : These results suggested that there are significant differences in DNA degradation by different food-processing methods . The likelihood of the large hpt gene fragments transfer from transgenic rice processed food to bacteria is reduced by food process .

Title: Molecular analysis of six segments of Tobacco leaf enation virus , a novel phytoreovirus from tobacco
Author: Picton A Potgieter C Rey ME .
Journal: Citation: V : ( ) P : Year: 2007 Type: ARTICLE
Literature: oryza Field: abstract Doc ID: pub17356907 Accession (PMID): 17356907
Abstract: Tobacco leaf enation virus ( TLEV ) is a putative member of the genus Phytoreovirus within the family Reoviridae . Previous western blot analysis of structural viral proteins ( apparent molecular weights of 93 kDa ; 58 kDa ; 48 kDa ; 39 kDa and 36 kDa ) associated with TLEV , isolated from infected tobacco in South Africa , suggested that these proteins may correspond to structural Wound tumor virus ( WTV ) proteins . To further establish the nature of this novel virus disease phenotype in tobacco , molecular characterization of six dsRNA components was undertaken . Full-length cDNA clones were obtained by an optimized modified single-primer amplification sequence-independent dsRNA cloning method . Results of this study revealed the conserved terminal sequence : 5GG ( U/C ) . . . UGAU 3 of segments S6-S12 , while adjacent to these conserved terminal sequences are imperfect inverted repeats ( 7-15 bp in length ) , both features being common to reoviruses . The complete nucleotide sequences of segments S5 ( 2 , 610 bp ) , S7 ( 1 , 740 bp ) , S8 ( 1 , 439 bp ) , S10 ( 1 , 252 bp ) , S11 ( 1 , 187 bp ) and S12 ( 836 bp ) were determined . Comparison of full-length nucleotide sequences with corresponding segments of other phytoreoviruses , Rice gall dwarf virus ( RGDV ) , Rice dwarf virus ( RDV ) and WTV has shown nucleotide and predicted amino acid identities within the range of 30-60% . TLEV consistently shows a higher identity to WTV than to other phytoreovirus species where sequence data is available . Each segment had a single predicted open reading frame encoding proteins with calculated molecular weights of S5 ( 90 . 6 kDa ) ; S7 ( 58 . 1 kDa ) ; S8 ( 47 . 7 kDa ) ; S10 ( 39 . 8 kDa ) ; S11 ( 35 kDa ) and S12 ( 19 . 5 kDa ) . The relatively low nucleotide and amino acid identity to other members of the genus demonstrates that TLEV is a novel phytoreovirus , distinct from the only other reported dicotyledenous-infecting WTV and is the first phytoreovirus reported to emerge in Africa .

Abstract: The complete nucleotide sequences of segments S2 , S3 , S5 , and S8-S11 of a Rice gall dwarf virus isolate from Guangxi Province , China , ( RGDV-GX ) were determined , completing the sequence of this isolate . The total genome ( 25 , 567nt ) was similar in organization to a recently reported Thailand isolate ( RGDV-T ) . A previously unreported second segment-specific repeat of 7 or 8nt was detected close to the 3-end of segments S3 , S5 , and S8 . The 3-UTR of RGDV-GX S4 was 139nt shorter than that of RGDV-T ; the insertion in RGDV-T contains a 34-nt inverted repeat , with the 3-terminus probably abolishing the expected stem loop structure .

Abstract: The 46K outer capsid protein encoded by RNA segment S8 and the 42K polypeptide , previously thought to be the segment S9-encoded structural protein , were isolated from a rice dwarf phytoreovirus purified preparation , and then analysed by peptide mapping and electroblot-ELISA . Staphylococcus aureus V8 protease peptide mapping patterns of the 42K and 46K proteins were similar . Two monoclonal antibodies ( MAbs ) , obtained after immunization with virus particles dissociated by 0 . 1% SDS , were each specific for both the 42K and 46K proteins . Furthermore , the MAbs bound common peptide fragments which were generated by digestion of the 42K and 46K proteins with V8 protease or proteinase K These results strongly suggest that the 42K protein is not a gene product of S9 but a product overlapping with the 46K outer capsid protein . Whether the two proteins are functionally distinct remains to be determined .

Abstract: The nucleotide sequences of DNAs complementary to the eighth ( S8 ) and the tenth ( S10 ) largest of the 12 genome segments of rice gall dwarf virus ( RGDV ) were determined . The S8 and S10 segments consist of 1578 and 1198 nucleotides , each with a single open reading frame extending for 1278 nucleotides from nucleotide 21 , and 960 nucleotides from nucleotide 22 , respectively . S8 encodes a polypeptide of 426 amino acids with an Mr of 47419 . The amino acid sequences of several peptide fragments of the major outer capsid protein reported as 45K were contained in the predicted polypeptide . This protein , renamed the 47K protein , showed high homology with the outer capsid proteins of rice dwarf virus ( RDV ) and wound tumour virus ( WTV ) ; there was 56 , 52 and 48% amino acid sequence identity between RGDV and WTV , RGDV and RDV , and RDV and WTV , respectively . S10 had the coding potential for a polypeptide of 320 amino acids with an Mr of 36 , 095 ( 36K protein ) , which exhibits 32% and 35% amino acid sequence identity with the predicted translation product of RDV S9 and the P9 capsid protein encoded by WTV S11 , respectively . The conserved terminal sequences 5 GG . . . GAU 3 which are present in all genome segments of WTV and RDV so far analysed , and in S9 of RGDV , were also found in RGDV S8 and S10 . This conserved sequence together with the segment-specific inverted repeats found in the terminal sequence of RGDV S8 and S10 are thus characteristic structures common to all three phytoreoviruses . The nucleotide sequence of the region surrounding the inverted repeats was more similar between RGDV and WTV than between RGDV and RDV .

Time points were : 3 , 6 , 12 , and 24 h after HIBD ( n=10 in each group at each time point ) . TanIIA was intraperitoneally given at 1 mug/g every 12 h . Fura-2AM was used to mark the fluorescent calcium probe and [ Ca ( 2+ ) ] ( i ) was measured by a Hitachi F-4500 Fluorescence Spectrophometer . Fluorescent immunohisotichemical study was used for the expression of phospho-NR1 S897 . RESULTS : ( 1 ) Compared with the normal control group , both the [ Ca ( 2+ ) ] ( i ) absolute number and ipsi-/contra-lateral ratio were increased at each time point with statistical significance ( P<0 . 05 ) . Compared with the HIBD group , the [ Ca ( 2+ ) ] ( i ) in the HIBD+ TanIIA group was decreased at each time point . At 24 h after HIBD , the ipsi-/contra-lateral ratio of HIBD+ TanIIA group was 24 . 9% less than that of HIBD group with statistical significance ( P<0 . 05 ) . ( 2 ) In the normal control group , abundant phospho-NR1 S897 positive cells were nicely distributed in the cortex Compared with the normal control group , at each time point , both the absolute number of phospho-NR1 S897 positive cells and the fluorescent intensity of phospho-NR1 S897 in the ipsilateral cortex of the HIBD group were decreased with statistical significance ( P<0 . 05 ) . Compared with the HIBD group , both the absolute number of phospho-NR1 S897 positive cells and the fluorescent intensity of phospho-NR1 S897 in the ipsilateral cortex of HIBD+ TanIIA were increased . There was significant difference at 3 and 12 h after the HIBD ( P<0 . 05 ) . CONCLUSION : TanIIA reduced the HIBD-caused down-regulation of phospho-NR1 S897 and the HIBD-caused [ Ca ( 2+ ) ] ( i ) elevation in the cortex The neuroprotective effect of TanIIA may be related to influencing NMDA receptor expression and decreasing intracellular free calcium aggregation .

Abstract: A new virus isolated from red raspberry plants and detected in the main production areas in northern Washington State , USA and British Columbia , Canada was fully sequenced and found to be a novel member of the family Reoviridae . The virus was designated as Raspberry latent virus ( RpLV ) based on the fact that it is symptomless when present in single infections in several Rubus virus indicators and commercial raspberry cultivars . RpLV genome is 26 , 128 nucleotides ( nt ) divided into 10 dsRNA segments . The length of the genomic segments ( S ) was similar to those of other reoviruses ranging from 3948 nt ( S1 ) to 1141 nt ( S10 ) . All of the segments , except S8 , have the conserved terminal sequences 5-AGUU----GAAUAC-3 . A point mutation at each terminus of S8 resulted in the sequences 5-AGUA----GAUUAC-3 . Inverted repeats adjacent to each conserved terminus as well as stem loops and extended pan handles were identified by analyses of secondary structures of the non-coding sequences . All segments , except S3 and S10 , contained a single open reading frame ( ORF ) on the positive sense RNAs . Two out-of-frame overlapping ORFs were identified in segments S3 ( ORF S3a and S3b ) and S10 ( ORF S10a and S10b ) . Amino acid ( aa ) alignments of the putative proteins encoded by the main ORF in each segment revealed a high identity to several proteins encoded by reoviruses from different genera including Oryzavirus , Cypovirus , and Dinovernavirus . Alignments of the polymerase , the most conserved protein among reoviruses , revealed a 36% aa identity between RpLV and Rice ragged stunt virus ( RRSV ) , the type member of the genus Oryzavirus , indicating that these two viruses are closely related . Phylogenetic analyses showed that RpLV clusters with members of the genera Oryzavirus , Cypovirus , Dinovernavirus and Fijivirus . These genera belong to the subfamily Spinareovirinae which includes reoviruses with spiked core particles ( turreted reoviruses ) . In addition , two nucleotide binding motifs , regarded as signature sequences among turreted reoviruses , were also found in RpLV P8 , suggesting that RpLV is a novel dicot-infecting reovirus in the subfamily Spinareovirinae .

Abstract: To obtain the P8 protein of Rice gall dwarf virus ( RGDV ) with biological activity , its outer coat protein gene S8 was expressed in Spodoptera frugiperda ( Sf9 ) insect cells using the baculovirus expression system . The S8 gene was subcloned into the pFastBac1 vector , to produce the recombinant baculovirus transfer vector pFB-S8 . After transformation , pFB-S8 was introduced into the competent cells ( E coli DH10Bac ) containing a shuttle vector , Bacmid , generating the recombinant bacmid rbpFB-S8 . After being infected by recombinant baculovirus rvpFB-S8 at different multiplicities of infection , Sf9 cells were collected at different times and analyzed by SDS-PAGE , Western blotting and immunofluorescence microscopy . The expression level of the P8 protein was highest between 48-72 h after transfection of Sf9 cells . Abstract: Recombinant proteins face major constraints along the plant cell secretory pathway , including proteolytic processing compromising their structural integrity . Here , we demonstrate the potential of protease inhibitors as in situ stabilizing agents for recombinant proteins migrating towards the leaf apoplast Genomic data for Arabidopsis , rice and Nicotiana spp . were assessed to determine the relative incidence of protease families in the cell secretory pathway . Transient expression assays with the model platform Nicotiana benthamiana were then performed to test the efficiency of protease inhibitors in stabilizing proteins targeted to the apoplast Current genomic data suggest the occurrence of proteases from several families along the secretory pathway , including A1 and A22 Asp proteases ; C1A and C13 Cys proteases ; and S1 , S8 and S10 Ser proteases . In vitro protease assays confirmed the presence of various proteases in N benthamiana leaves , notably pointing to the deposition of A1 and S1-type activities preferentially in the apoplast Accordingly , transient expression and secretion of the A1/S1 protease inhibitor , tomato cathepsin D inhibitor ( SlCDI ) , negatively altered A1 and S1 protease activities in this cell compartment , while increasing the leaf apoplast protein content by approximately 45% and improving the accumulation of a murine diagnostic antibody , C5-1 , co-secreted in the apoplast SlCYS9 , an inhibitor of C1A and C13 Cys proteases , had no impact on the apoplast proteases and protein content , but stabilized C5-1 in planta , presumably upstream in the secretory pathway . These data confirm , overall , the potential of protease inhibitors for the in situ protection of recombinant proteins along the plant cell secretory pathway .

Abstract: In a previous study , we reported the ecological significance of uncultured bacterial group U2 in the rumen . In this study , the involvement of a recently-cultured group U2 bacterium , strain R-25 , in fiber digestion was tested in coculture with the fibrolytic bacterium Fibrobacter succinogenes S85 . Dry matter ( DM ) digestion , growth and metabolites were examined in culture using rice straw as the carbon source . Although strain R-25 did not digest rice straw in monoculture , coculture of strain R-25 and F succinogenes S85 showed enhanced DM digestion compared to that for F succinogenes S85 monoculture ( 36 . 9+/-0 . 6% vs 32 . 8+/-1 . 3% , P < 0 . 05 ) . Growth of strain R-25 and production of the main metabolites , D-lactate ( strain R-25 ) and succinate ( F succinogenes S85 ) , were enhanced in the coculture . Enzyme assay showed increased activities of carboxymethylcellulase and xylanase in coculture of strain R-25 and F succinogenes S85 . Triculture including strain R-25 , F succinogenes S85 and Selenomonas ruminantium S137 showed a further increase in DM digestion ( 41 . 8+/-0 . 8% , P < 0 . 05 ) with a concomitant increase of propionate , produced from the conversion of D-lactate and succinate . These results suggest that the positive interaction between strains R-25 and F succinogenes S85 causes increased rice straw digestion . ( c ) 2012 Federation of European Microbiological Societies . Abstract: The nucleotide sequence of DNA complementary to rice ragged stunt oryzavirus ( RRSV ) genome segment 8 ( S8 ) of an isolate from Thailand was determined . RRSVS8 is 1914 bp in size and contains a single large open reading frame ( ORF ) spanning nucleotides 23 to 1810 which is capable of encoding a protein of M ( r ) 67 , 348 . The N-terminal amino acid sequence of a approximately 43K virion polypeptide matched to that inferred for an internal region of the S8 coding sequence . These data suggest that the 43K protein is encoded by S8 and is derived by a proteolytic cleavage . Predicted polypeptide sizes from this possible cleavage of S8 protein are 26K and 42K . Polyclonal antibodies raised against a maltose binding protein ( MBP ) -S8 fusion polypeptide ( expressed in Escherichia coli ) recognised four RRSV particle associated polypeptides of M ( r ) 67K , 46K , 43K and 26K and all except the 26K polypeptide were also highly immunoreactive to polyclonal antibodies raised against purified RRSV particles . Cleavage of the MBP-S8 fusion polypeptide with protease Factor X produced the expected 40K MBP and two polypeptides of apparent M ( r ) 46K and 26K . Antibodies to purified RRSV particles reacted strongly with the intact fusion protein and the 46K cleavage product but weakly to the 26K product . Furthermore , in vitro transcription and translation of the S8 coding region revealed a post-translational self cleavage of the 67K polypeptide to 46K and 26K products . These data indicate that S8 encodes a structural polypeptide , the majority of which is auto-catalytically cleaved to 26K and 46K proteins . The data also suggest that the 26K protein is the self cleaving protease and that the 46K product is further processed or undergoes stable conformational changes to a approximately 43K major capsid protein .

Abstract: Sequence determination of full-length cDNA clones of genomic segments 7-10 ( S7-S10 ) of oat sterile dwarf fijivirus ( OSDV ) revealed that the 5 and 3 ends of the plus strands of these segments had the same conserved terminal sequences , 5 AACGAAAAA and UUUUUUUAGUC 3 . These sequences are similar , but not identical , to the conserved terminal nucleotide sequences of the genomic segments of rice black streaked dwarf fijivirus ( RBSDV ) and maize rough dwarf fijivirus ( MRDV ) . The coding strands of S7 and S10 each contained two large nonoverlapping open reading frames ( ORFs ) , as do RBSDV S7 and S9 , MRDV S6 and S8 and Nilaparvata lugens reovirus ( NLRV ; a putative member of Fijivirus ) S9 . These results strongly suggest that the dicistronic nature of certain genomic segments is characteristic of fijiviruses . Computer analyses revealed sequence homology between RBSDV S7 ORF2 , MRDV S6 ORF2 and OSDV S7 ORF2 , suggesting that this protein is conserved among plant fijiviruses . No counterparts were found in the genome of NLRV , which is a nonphytopathogenic insect reovirus . Furthermore , phylogenetic trees derived from multiple sequence alignments of each of the homologous proteins from OSDV , RBSDV , MRDV and NLRV suggest that NLRV did not evolve from either Fijivirus group 2 ( RBSDV and MRDV ) or group 3 ( OSDV ) .

Abstract: The proteins encoded by rice black streaked dwarf fijivirus ( RBSDV ) genomic segments 7-10 ( S7-S10 ) were characterized . Open reading frames ( ORFs ) from these segments were expressed as fusion proteins in Escherichia coli . Antibodies raised against the expressed products were used as probes to determine whether the viral ORFs encode structural proteins . In Western blots , antibodies to the expressed S8 and S10 products reacted with a core capsid ( 65 kDa ) and a major outer capsid ( 56 kDa ) protein , respectively , while none of the antibodies to S7 and S9 products reacted with structural proteins . Antisera to RBSDV S7 ORF1 and S9 ORF1 each detected a single protein of the predicted size in total protein extracts from infected rice plants and viruliferous Laodelphax striatellus . Immunoelectron microscopy revealed that antibodies to RBSDV S7 ORF1 and RBSDV S9 ORF1 reacted with tubular structures and viroplasm , respectively , in sections of both infected maize plants and viruliferous L striatellus .

Immunofluorescence microscopy showed that P8 protein of RGDV formed punctate structures in the cytoplasm of Sf9 cells .

Antisera to ORF2 of S7 and S9 failed to detect any proteins in the infected it issue using either Western blotting or immuno-electron microscopic techniques .
